Aging is an inevitable consequence of life for nearly all organisms. It mostly reflects the outcome of complicated interactions between genetic factors along with the accumulation of a variety of deleterious stochastic changes over time. Thus, aging is an adaptive process, not caused by a single factor or process, but it is rather a multifactorial procedure modulated by the interplay among genetic and environmental factors. Since aging is a major risk factor in connection with a variety of human disorders, it is increasingly becoming a central topic in biochemical and medical research. The plethora of theories on aging – some of which have been discussed for decades – are neither isolated nor contradictory but instead can be connected in a network of pathways and processes at the cellular and molecular levels. This book summarizes the most prominent and important approaches, focusing on telomeres, DNA damage and oxidative stress as well as on the possible role of nutrition, the interplay between genes and environment (epigenetics) and intracellular protein homeostasis and introduces some genes that have actually extended life spans in animal models. Linking these different determinants of aging with disease, this volume aims to reveal their multiple interdependencies. We see that there is no single “perfect” theory of aging and that instead it is possible to define what the authors call the molecular aging matrix of the cell. A better knowledge of its key mechanisms and the mutual connections between its components will lead to a better understanding of age-associated disorders such as Alzheimer’s disease.

As we approach the twenty-first century, aging is likely to emerge as a leading issue for biological and biomedical scientists. The elucidation of the underlying mechanisms of aging may constitute the most effective approach to the prevention or postponement of major geriatric disorders. An understanding of why individual subjects vary in their susceptibilities to these disorders will be of special significance to future medicine. To further this understanding, a unique dialogue was initiated between biologists, geneticists, biochemists, pathologists, immunologists, physicians, and other biomedical scientists concerned with the molecular mechanisms of aging, which is reflected in this volume. The authors consider model systems based on yeast, simple fungi, roundworms, fruitflies, mice, and humans. They review the current status of the evolutionary theory of aging, of oxidatively modified proteins as markers of aging, the role of mitochondrial and nuclear DNA damage in aging, and the pathogenic mechanisms in common geriatric disorders such as Alzheimer?s disease, Parkinson?s disease, atherosclerosis, and cancer. The reports summarize their debate on key issues: How can research on diverse model systems contribute to the understanding of the genetic basis of aging and longevity? Do research findings from diverse model systems support oxidative damage theories of aging? Does research on mitochondria in diverse model systems provide at least one unifying mechanism of aging? Do common underlying mechanisms of aging contribute to the pathogenesis of major geriatric disorders? Goal of this Dahlem Workshop: to examine molecular processes underlying genetic and physiological changes that occur with normal aging and geriatric disorders.

Aging is a degenerative process that causes a time-dependent deterioration of virtually every biological system in the majority of species. Age is the primary risk factor for many human diseases, including the top causes of death modern societies. Developing treatments to slow the aging process has the potential increase human life span and simultaneously prevent or improve outcomes in countless diseases. Studying aging in mammals is challenging due to the relatively high longevity of most mammalian species and the costs associated with maintaining populations of mammals in the laboratory for their entire life span. The invertebrate organisms Saccharomyces cerevisiae, Caenorhabditis elegans, and Drosophila melanogaster have emerged as central models in aging due to relatively short life spans, ease of maintenance in the laboratory, well characterized genetics, and the availability of a wide range of genetic and biochemical tools. By focusing on genetic pathways and interventions that influence longevity in a similar manner across these evolutionarily divergent species, we can gain insight into the biology of aging in mammals. The application of genome-scale techniques in aging research has started to define the range of genetic and environmental factors involved in longevity determination, and the high degree of intercommunication between these factors. This dissertation reviews current progress toward identifying and understanding conserved longevity interventions and presents several current lines of investigation aimed both at developing tools for analyzing the complex interactions between aging factors and at probing the mechanism of action of specific longevity interventions.
